The rifling is cut with a left-hand, 1-in-14-inch twist across six grooves, a relatively gentle rate that stabilizes the wide range of bullet weights commonly loaded in .357 Magnum, from light 110-grain defensive rounds to heavy 180-grain hunting projectiles. The adjustable rear sight works in conjunction with a user-interchangeable red ramp front sight, allowing the shooter to zero precisely for a chosen load rather than splitting the difference across a fixed sight picture.

The frame, barrel, and cylinder are all stainless steel. Colt reinforced the area beneath the rear sight with significantly more material than the previous-generation Python carried, addressing a known stress point.
